Rocco Mastroserio was born in Barre, Italy, but was raised in the USA. His earliest comic book work was as an inker on crime and western titles by Avon Comics and Prize Comics in the early 1950s. He also contributed to ACG's 'Adventures into the Unknown' and 'Operation: Peril', Key Publications' 'Mister Mystery' and the Harvey horror anthologies 'Black Cat Mystery', 'Chamber of Chills', 'Tales of Horror', and 'Tomb of Terror'.

He has mainly worked for Charlton Comics. He did his first work for the satirical comic book 'Eh!', and continued to draw for the company's many western, adventure, mystery and science fiction titles throughout the 1950s and 1960s, such as 'Billy the Kid', 'Cheyenne Kid', 'Fightin' Army', 'Fightin' Navy', 'Kid Montana', 'Space Adventures', 'Wyatt Earp' and 'Strange Suspence Stories'. He also inked most 'Captain Atom' stories pencilled by Steve Ditko, and is the co-creator of the Charlton superhero 'Mercury Man'. In the late 1960s, he illustrated several stories written by Archie Goodwin for the Warren horror magazines Creepy and Eerie.
